No. The Squid process requires exclusive access to the cache
directories.

What you can do, is to have two squids on the same machine, where one is
parent/neighbour to the other (of course running on different ports, and
different cache directories). This might be useful if you want to test a
new/patched version before switching. The test version requires very a
very small cache, but benefits from the large cache by querying the
standard squid process.
