Squid as such only supports a single authentication helper per
authentication scheme, but it is trivial to use a little glue between
Squid and the authentication helpers, selecting which backend
authentication helper to use based on the username.

You cannot select which backend authentication method to use based on
the site or URL visited. You can only select authentication method based
on the entered user name (or password)

For a silly example of such authentication method glue, search for Open2
in the squid-users archives.
