Squid starts replacing old content with new. As needed. You do not have
to clear anything.

Just ensure that the space configured is actually well less than the
available disk space. There is a journal 'swap.state' which Squid keeps
adding to. Regular "squid -k rotate" (daily / weekly) keep this journal
and the logs as well clean and small.
