This rule is exactly the culprit!!
You need to route the traffic towards the squid machine and not do
redirection\nat towards a specific port.
Just route...
Probably on the squid box you will see the microtik ip as src ip for the
request which is wrong..
Consider to also look at this:

Which is another thing but should also be a nice idea.
