Then you have major problems with your setup. Squid absolutely MUST be
capable to function as a standard proxy before being configured as a
transparent proxy. If it won't work in non-transparent mode then there
is no chance it will work in transparent mode.

You haven't read the FAQ on transparent caching with Squid.

httpd_accel_host virtual
httpd_accel_uses_host_header on

and also recommended to allow both transparent and proxy mode at the
same time:
httpd_accel_with_proxy on
