Traffic flow with SQUID implementation

Dear all,
I'm planning to implement SQUID in a local ISP but am not too sure of how
exactly the flow of packets will take place. Consider the basic network
scenario illustrated below:
-------------------- --------------
| Customer networks | --> | ISP Router | ----> INTERNET
-------------------- --------------
                                            |(Ethernet)
                                            |
                                SQUID CACHE

Does the ISP router need to re-route all HTTP traffic transparently to the
cache server (redirect port 80) and have all the client PCs browsers
configured to point to the SQUID as a proxy server using port 3128?
Could you please describe the flow of traffic in this simplest of scenarios?
Went through the FAQs etc but couldn't find a clear explanation.
