Normally you should not need to as Squid automatically will delete
content from the cache to make space for new objects.

But if you from some reason should need to, just delete some files.
Squid will recover gracefully when it finds that the file is missing
with only a small complaint to cache.log..

Or you could use the PURGE method to selectively delete known objects
from the cache.
