I am serving about 1500 users on an ex compaq proliant ML370 box
which is PIII based and 1Gb Ram running
redhat Linux 6.2. This machines make me very happy.
During office hours squid is serving an average of 30url's/sec.
But this hardware has more potential and I think the box
could easily handle 60 reqs/sec or more.

Cache size (disk space) will depend on your end
 user's 'web access profile'.
For instance try to have an idea about the amount of Megs transferred
per week. This could be an adequate indicator about the needed cache
size.

I have 11Gb's going 'through squid' each day (more or less).
I configured a 40Gb cache, giving me an average of 50% hitrate
 nowadays.
