 Normally yes, if squid is stopped first.

 If adequate cache dir's are specified in squid.conf , then
 the cache can be 'full' in terms of cache sizes specified.
 Squid maintains these conditions by itself and performs
 regular cleanups of old objects (simply stated).

 There is no need for intervention, unless of course this
 would relate to filesystems overflowing , etc.
