You do not need to restart Squid. A "squid -k reconfigure" is
sufficient and much less intrusive on the service.
I am not aware of a acl helper doing exacly this, but it should not be
hard to write one I suppose. But some thought may be needed in how to
index "sites" into a relational databases if you by "sites" mean
something more than only fully qualified host names..
