the only way I see is to remove one of cache_dirs off squid.conf and
restart squid, remove the data on the disk and increase second cache_dir
to whole disk. You'll loose its contents of course.

I would try bigger number, even if it's hard to decide the best setting.

I currently have 256KB (but probably will decrease it a bit to see if it
helps the memory hit ratio), for your setup I'd probably try 64-128KB.
